The composition and distribution of photoperiod, vernalization and dwarf alleles in parents of two-line hybrid wheat

Abstract:To clarify the composition and distribution of photoperiod, vernalization and dwarf alleles in the parent
materials of two-line hybrid wheat, a total of 245 parent materials of two-line hybrid wheat were detected by KASP (kompetitive allele specifific PCR) markers for photoperiod, vernalization and dwarf genotypes. The results indicated that Ppd-A1b, Ppd-D1b and Ppd-B1b were the three genotypes of photoperiod gene in the tested materials, and the frequency of Ppd-A1b and Ppd-D1b were 100% both in restorer and sterile lines, while Ppd-B1b was 87.98% and 91.67% in restorer and sterile lines, respectively. The genotype of vernalization gene was vrn-A1 in the tested materials, the frequency of which was 98.71% and 100% in restorer and sterile lines, respectively. The dwarf genes Rht-B1a and Rht-D1a were present in the restorer lines with the frequency of 98.28% and 94.42%, respectively. Rht-B1a and Rht-D1b were observed in the sterile lines with the frequency of 66.67% and 100%, respectively. This study has signifificant value for further development of adaptive genetic improvement of hybrid wheat. In addition, KASP assays could greatly improve speed and effificiency of the two-line hybrid wheat breeding programs by identifification and screening of genetic materials with more valuable allelic genes.
